First-line treatment: Patients with metastatic NSCLC harboring EGFR exon 19 deletions or exon 21 L858R mutations (detected by an FDA-approved test).
Second-line treatment: Patients with metastatic EGFR T790M mutation-positive NSCLC whose disease has progressed after previous EGFR tyrosine kinase inhibitor therapy.

✔️ Mechanism of Action: Tasso 80

Irreversible inhibition of mutant forms of EGFR (T790M, L858R, exon 19 deletions).
Two active metabolites (AZ7550 and AZ5104) contribute to its activity.
Targets HER2, HER3, HER4, and other kinases at clinically relevant levels.

✔️ Dosage and Administration of Tasso 80

Standard Dose: 80 mg orally once daily.
Administration Tips:
- Can be taken with or without food.
- Tablets can be dispersed in non-carbonated water for easier swallowing.
- Avoid crushing or heating the tablets.

Common (≥20%):

Diarrhea, rash, dry skin, nail changes, stomatitis, fatigue, and reduced appetite.

Serious:

Interstitial lung disease (ILD)/Pneumonitis (3.9%, rare fatal cases).
QTc prolongation: Monitor ECG and electrolytes.
Cardiomyopathy: Regular LVEF assessments are recommended.

Take one 80 mg tablet once daily, at the same time each day. Swallow the tablet whole without crushing, splitting, or chewing. For patients with swallowing difficulties, disperse the tablet in 50 mL of non-carbonated water, stir without crushing, and drink immediately. Rinse the glass with another ½ glass of water and drink. The dispersed solution can also be administered via a nasogastric tube, ensuring the tube is flushed with water after administration.

For mild to moderate renal impairment (CrCl 30-89 mL/min): No dose adjustment is needed. For severe renal impairment or end-stage renal disease (CrCl <30 mL/min): There is no recommended dose
